摘要

目的:观察沉默穴位局部缝隙连接蛋白(connexin43

Cx43)的表达对针刺效应的影响

探讨针刺治疗原发性痛经的机制。方法:以催产素造成大鼠原发性痛经模型

应用RNA干扰(RNAi)技术沉默穴位局部Cx43的表达。将50只SD雌性大鼠分为正常组(N)、模型组(M)、针刺组(A)、针刺+干扰组(A+I)、针刺+干扰对照组(A+IC)。应用RT-PCR方法检测大鼠子宫的催产素受体(oxytocin receptor

OTR)、加压素受体(vasopressin receptor

VPR)的mRNA表达情况;放免法和ELISA检测血浆中前列腺素E2(PGE2)和PGF2α的含量。结果:①A组和A+IC组的大鼠扭体次数(9.43±3.87

10.28±4.23)明显低于M组和A+I组(15.43±5.13

17.00±3.87)

潜伏期(12.43±3.46

11.00±3.65)min高于M组(7.57±1.99)min和A+I组(8.43±2.57)min(P<0.05)。②A+I组穴位处的Cx43 mRNA和蛋白表达低于N组(P<0.05)。③A组和A+IC组的OTR、VPR的mRNA表达较M组和A+I组降低

且差异有统计学意义(均P<0.05);M组和A+I组差异无统计学意义(P>0.05)。④A组和A+IC组的PGE2升高

PGF2α降低

较M组差异有统计学意义(P<0.05)。结论:沉默穴位局部Cx43能有效抑制针刺效应

通过降低痛经大鼠子宫内膜OTR、VPR及调节前列腺素合成系统

可能是针刺治疗原发性痛经的作用机制之一。
