摘要

目的:研究苦参素“足三里”注射对2-乙酰氨基芴(2-AAF)诱发大鼠实验性肝癌的作用

并探讨其抑制肝癌细胞增殖的机制。方法:将肝癌大鼠随机分为4组:正常对照(N)组、模型对照(M)组、苦参素腹腔注射(OMip)对照组、苦参素小剂量“足三里”(OMZSL)穴注组。实验至12w末

取血测定血清丙氨酸氨基转移酶(ALT)、谷草转氨酶(AST)及γ-谷氨酰转肽酶(γ-GT)水平;RT-PCR法检测肝癌组织中细胞周期素D1(cyclinD1)、细胞周期蛋白依赖激酶4(CDK4)mRNA的表达。结果:OMip组和OMZSL小剂量组大鼠肝表面癌结节数低于M组

能明显降低肝癌大鼠血清ALT、AST及γ-GT水平(P<0·01)

明显抑制肝癌组织中cyclinD1、CDK4mRNA的表达(P<0·01)。结论:OMip和OMZSL小剂量给药能治疗或延缓2-AAF诱发大鼠肝癌发生的作用

其部分机制可能通过降低肝癌大鼠血清ALT、AST和γ-GT水平保护肝细胞

从而间接发挥治疗肝癌作用;通过抑制cyclinD1、CDK4mRNA的表达

从而诱导细胞周期阻滞

抑制肝癌细胞过度增殖。
