摘要

目的:观测逆灸对力竭运动大鼠心肌组织中一磷酸腺苷活化的蛋白激酶(AMPK)、哺乳动物雷帕霉素靶蛋白(mTOR)的影响

探讨逆灸抗力竭运动心肌损伤的作用机制。方法:将18只雄性SD大鼠随机分为空白组、力竭组和逆灸组

每组6只。空白组大鼠每天被操作者用手轻按于操作台上

套上特制鼠套

保持5min

不进行任何干预;力竭组大鼠前10d不做处理

逆灸组取"足三里""关元"进行干预

每穴每日灸5壮

连续10d

第11d力竭组和逆灸组大鼠力竭游泳后取材

并记录两组大鼠达到力竭所用时间。采用HE染色观察各组大鼠心肌组织炎性反应;比色法检测血清中乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;免疫抑制法检测血清中肌酸激酶同工酶(CK-MB)活性;Elisa检测血清中肌钙蛋白T(cTnT)含量;Western blot测定心肌组织中一磷酸腺苷活化的蛋白激酶α2(AMPKα2)、哺乳动物雷帕霉素靶蛋白(mTOR)表达。结果:逆灸组较力竭组到力竭状态所需时间延长[(4 831.17±689.88)s vs(3 509.50±1 232.49)s

心肌组织结构清楚

炎细胞浸润减轻;与空白组比较

力竭组血清LDH、CK-MB、cTnT升高(均P<0.05);MDA含量升高

SOD活性下降(均P<0.05);心肌组织AMPKα2表达升高、mTOR下降(均P<0.05)。与力竭组比较

逆灸组血清LDH、CK-MB、cTnT下降(均P<0.05);MDA含量下降

SOD活性升高(均P<0.05);心肌组织AMPKα2表达升高、mTOR下降(均P<0.05);AMPKα2与mTOR呈负相关(r=-0.764

P<0.05)。结论:逆灸可能通过调控AMPK/mTOR表达诱导相应的信号级联反应对抗一次性力竭运动大鼠心肌组织损伤。
