摘要

目的:探讨头穴丛刺对阿尔茨海默病(Alzheimer’s disease

AD)大鼠海马区乙酰胆碱转移酶(ChAT)、乙酰胆碱酯酶(AchE)表达的影响。方法:将60只Wistar大鼠随机分为假手术组、模型组、药物组、丛刺组

每组15只。模型组、药物组、丛刺组采用Aβ1-42向大鼠双侧海马CA1区定向注射诱导阿尔茨海默病(AD)模型

假手术组给予双侧海马区注射等量蒸馏水。药物组给予多奈呱齐(安理申)灌胃作为基础治疗

每日1次

共治疗4周;丛刺组在药物组的基础上加头穴丛刺治疗

每日1次

每周休息1d

共干预4周;假手术组、模型组给予常规喂养。干预后利用Morris水迷宫检测大鼠学习记忆能力;HE染色观察大鼠海马大体结构的变化;免疫组化观察大鼠海马区Ch AT、Ach E阳性表达情况。结果:与假手术组比较

模型组大鼠的逃避潜伏期延长(P<0.01)

第二象限百分比和跨越平台次数减少(均P<0.01);与模型组比较

丛刺组、药物组大鼠的逃避潜伏期缩短(P<0.05

P<0.01)

第二象限百分比和跨越平台次数增加(P<0.05

P<0.01);与药物组比较

丛刺组大鼠的逃避潜伏期缩短(P<0.05)

第二象限百分比和跨越平台次数增加(均P<0.05)。与假手术组比较

模型组ChAT表达下降(P<0.01)

Ach E表达上升(P<0.01);与模型组相比

药物组Ch AT差异无统计学意义(P>0.05)

AchE表达下降(P<0.05)

丛刺组海马区ChAT表达增加(P<0.05)

AchE表达下降(P<0.01);与药物组相比

丛刺组ChAT表达上升(P<0.05)

AchE表达下降(P<0.05)。结论:头穴丛刺辅助治疗AD的机制之一可能是上调海马区ChAT活性及下调AchE活性

头穴丛刺联合安理申疗效更佳。
