摘要

目的:观察长时间不同浓度艾烟环境对大鼠嗅觉功能的影响

为艾灸过程产生的艾烟安全性研究提供实验依据。方法:将40只SD大鼠随机分为正常对照组、低浓度艾烟组(低烟组)、中浓度艾烟组(中烟组)和高浓度艾烟组(高烟组)

每组10只。艾烟组大鼠分别放入3个不同艾烟浓度有机玻璃艾灸箱内进行每日2次、每次4 h的艾烟烟熏干预

持续90 d。实验前及实验中对大鼠一般状态进行评估;干预结束后

通过双瓶实验(TBE)对大鼠进行嗅觉评估

采用HE染色观察大鼠鼻黏膜组织形态、TUNEL法检测大鼠鼻黏膜嗅上皮细胞凋亡情况

采用ELISA法检测大鼠血清白介素1(IL-1)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)的含量。结果:艾烟烟熏后期(干预45～90 d)

中烟组、高烟组大鼠行为活动较正常对照组大鼠减弱

且对刺激反应强烈

精神状态较差;干预结束后

中烟组、高烟组大鼠醋水混合液饮用率均高于正常对照组、低烟组(P<0.01);中烟组、高烟组大鼠鼻黏膜层次结构不清、排列紊乱

有坏死

炎性细胞浸润严重;中烟组、高烟组大鼠鼻黏膜嗅上皮凋亡细胞数多于正常对照组、低烟组(P<0.01)

高烟组多于中烟组(P<0.01);低烟组、中烟组、高烟组大鼠血清IL-1、IL-6、TNF-α含量高于正常对照组(P<0.01)

且中烟组、高烟组高于低烟组(P<0.01)

高烟组高于中烟组(P<0.01)。结论:长期暴露在低、中、高浓度艾烟环境中均会引起大鼠鼻黏膜组织病理改变

导致血清IL-1、IL-6、TNF-α含量升高

并且中、高浓度艾烟环境会对嗅觉功能产生一系列损害

降低大鼠嗅觉灵敏度。
