摘要

目的:观察艾灸"足三里"对溃疡性结肠炎(UC)模型小鼠不同时间段结肠远端、中段、近端黏膜损伤及结肠远端黏膜层降钙素基因相关肽(CGRP)阳性神经纤维表达的影响。方法:将51只C57BL/6N小鼠随机分为7 d对照组(8只)、7 d模型组(7只)、7 d艾灸组(7只)、14 d对照组(6只)、14 d模型组(14只)和14 d艾灸组(9只)。模型组和艾灸组均予2%葡聚糖硫酸钠(DSS)溶液自由饮用7 d建立UC模型。造模3 d后

7 d艾灸组及14 d艾灸组予艾灸"足三里"干预

每天1次

每次10 min

分别干预5、12 d。采用HE染色法观察各组小鼠结肠组织形态

计算结肠远端、中段、近端黏膜损伤长度百分比。采用免疫荧光法观察各组小鼠结肠远端、中段、近端损伤部位黏膜层神经纤维及结肠远端黏膜层CGRP阳性神经纤维的表达。结果:造模后小鼠出现多处黏膜损伤

表现为上皮层消失、隐窝结构异常或消失。与7 d对照组比较

7 d模型组小鼠结肠长度更短(P<0.001)

结肠全段、远端及中段黏膜损伤长度百分比增加(P<0.001);结肠远端、中段、近端损伤部位黏膜层神经纤维阳性表达及远端黏膜层CGRP阳性神经纤维表达增加(P<0.001

7 d艾灸组小鼠结肠远端及中段损伤部位黏膜层神经纤维阳性表达及远端黏膜层CGRP阳性神经纤维表达减少(P<0.05)。与14 d对照组比较

结肠全段黏膜损伤长度百分比增加(P<0.001)。与14 d模型组比较

14 d艾灸组小鼠结肠长度更长(P<0.05)

结肠全段黏膜损伤长度百分比减少(P<0.05)。结论:艾灸"足三里"可减少UC模型小鼠结肠黏膜层神经纤维及CGRP阳性神经纤维表达

从而改善结肠黏膜损伤程度。
